研究方向:
1.腺病毒疫苗及载体(Adenovirus vaccines and vectors)
近年来人3、4、 7、14、55型腺病毒感染在美洲、欧洲、亚洲,特别是在我国幼儿园、学校以及军队中的感染频频暴发,目前无安全的腺病毒疫苗,也无特效的治疗方法,因此迫切需要进行相关的疫苗研究。研制开发价格低廉、安全有效、具有我国自主知识产权的腺病毒疫苗,经济效益和社会效益巨大,应用前景广阔。本研究利用人5型复制缺陷型腺病毒载体,构建同时表达其他型别腺病毒六邻体蛋白的复制缺陷型重组腺病毒疫苗候选株,为今后腺病毒疫苗的应用研究奠定基础。同时,鉴于目前尚无合适的腺病毒动物感染模型,我们拟建立一种新的腺病毒小动物感染模型,以推动腺病毒疾病体内研究的飞速发展。目前该研究受国家自然科学基金、广东省自然科学基金和珠江科技新星项目支持。
2.腺病毒分子流行病学、基因组学、进化(Genomics, molecular epidemiology and evolution of adenoviruses)
近年来中国频频出现腺病毒的感染和暴发,大量的报道表明,腺病毒开始出现越来越多的基因组变异与重组,引起暴发的变异重组株显示出逃避机体免疫的能力。因此,本研究计划对我国当前流行的腺病毒进行全基因组测序和进化分析,构建腺病毒体内/体外基因组重组的模型,找出其流行及基因组变异重组的规律,并预测腺病毒的进化趋势,为今后腺病毒功能基因组学研究、开发腺病毒疫苗以及我国腺病毒疾病的防控提供科学依据。目前该研究受国家自然科学基金面上项目、广东省高等学校优秀青年教师培养计划支持。
RESEARCH INTERESTS:
My research interests are in the areas of genomics, epidemiology, and vaccines for human adenoviruses, especially for those causing children diseases, such as pink eye, pneumonia and other acute respiratory disease. Recently, there have been more and more human acute respiratory disease outbreaks in Chinese schools and militaries caused by adenoviruses. Homologous recombination appears to play a major role in generating new adenoviruses. So the comprehensive analysis of whole-genome sequences for these existing and emergent adenoviruses is an important approach to identify the homologous recombination and transmission chains in adenovirus evolution, which may lead to new and more severe human infections. In addition, adenovirus infections are very common in children aged 0.6-3, which pose a threat to their health. As a result, I am also interested in the development of effective vaccine candidates using new strategies for those pandemic adenoviruses, as well as the construction of optimal animal models.
